Immobilization Resins
Activated immobilization resins from ProteoChem can aid your research by facilitating the production of your immobilized ligand of choice. Whether you have proteins such as enzymes, antibody or simple peptides, ProteoChem has the activated gel for you.
1. Amino-Reactive Resin
Amino-Reactive Resin is an activated amine reactive agarose resin that is a great choice for immobilizing
antibody, protein, or peptides in order to make affinity resins for column chromatography that can purify
your protein of choice. Covalently immobilize 1 to 10 mg of antibody/ml of coupling resin using Sodium
cyanoborohydride as the reductant.
Amino-Reactive Coupling Resin is a chemically modified resin with aldehyde molecules immobilized on 6% crosslinked agarose. The bead size is 45 to 165 um. Primary amines, as found in lysine residues of proteins and peptides, can be immobilized and covalently bound to the activated agarose resin to create your specific affinity chromatography resin.
2. Sulfhydryl-Reactive Resin
Sulfhydryl-Reactive Resin coupling gel is an immobilized iodoacetyl resin
(Thiol Reactive Agarose) with a 12-atom spacer arm. It is a great choice for
immobilizing cysteine containing peptides or protein in order to make your
very own affinity resin and purify your antibody or protein of choice.
Sulfhydryl-Reactive Resin is supplied as a 50% slurry.
Sulfhydryl-Reactive Resin is a chemically modified agarose resin with iodoacetyl molecules immobilized on 6% crosslinked agarose. The bead size is 45 to 165 um. Sulfhydryl (thiol; -SH) containing peptides (0.5 -1 mg/mL resin) and protein (~ 1-10 mg/mL resin) can be covalently bound to Sulfhydryl-Reactive Agarose resin to create your specific affinity chromatography resin.

3. Carbohydrate-Reactive Resin
Carbohydrate-Reactive Resin (hydrazide-agarose, HYD-Agarose) is a coupling resin
is used for immobilizing glycones and other glycated molecules, including
glycoproteins and antibodies through their carbohydrates (compare to Thermo
Scientific Carbolink or Pierce Carbolink *). Carbohydrate-Reactive Resin is
supplied as a 50% slurry (15mL resin = 30mL slurry).
Carbohydrate-Reactive Agarose Resin is a chemically modified agarose gel with a hydrazide group immobilized on 6% crosslinked agarose. The bead size is 45 to 165 um. It is ideal for for coupling small ligands containing reactive formyl groups or to immobilize proteins that contain oxidizable carbohydrates. For glycoproteins, the coupling reaction takes place by oxidizing the carbohydrate group on your glycoprotein with Sodium meta-Periodate and combining the oxidized glycoprotein with the resin. The spacer arm in this resin enables increased coupling efficiency by reducing sterics.

4. Carboxyl-Reactive Resin
Carboxyl-Reactive Resin (Immobilized DADPA coupling resin; DADPA Agarose) is a useful carboxyl coupling resin with an 8-atom spacer arm and a free primary amine (-NH2). ProteoChem's Carboxyl-Reactive Resin is DADPA-activated crosslinked 6% beaded agarose that can be used to immobilize carboxylic acid (-COOH) containing compounds or proteins for generating affinity columns through the carboxyl groups when used in combination with EDC-HCl (EDAC-HCl). Carboxyl-Reactive Resin coupling gel is supplied as a 50% slurry (15 mL resin = 30 mL slurry).
Support : Diaminodipropylamine (DADPA) is immobilized on 6% crosslinked beaded agarose gel.
Activation Level : ≥ 16 μmol amine/ml of resin.
5. Immobilized Pepsin
Immobilized Pepsin resin (Pepsin Agarose) is commonly used to create F(ab’)2 fragments from antibodies. By immobilizing pepsin autolysis and protease contamination is essentially eliminated. Immobilized Pepsin gel hydrolyzes protein non-specifically at pH 1.0 to 3.0, with an optimum activity at pH 1.0. ProteoChem's highly active pepsin is covalently bound to crosslinked 6% beaded agarose resin and its specific activity is > 6,100 units/ml resin (one unit is defined as releasing 0.001 A280 as TCA hydrolyzed/min. at 37°C). ProteoChem's Immobilized Pepsin is packaged as a slurry in buffer at pH 4.5 containing 50% glycerol, 100 mM Sodium Acetate, 0.05% NaN3.
Pepsin protease cleaves non-specifically with some preference for tyrosines and phenylalanine (aromatic residues), although it generally does not cleave at alanine, glycine or valine residues. Recommended conditions are pH 1.0-4.5, 20°C to 37°C.

6. Immobilized Chymotrypsin
Immobilized Chymotrypsin Resin (Chymotrypsin Agarose) is used to preferentially hydrolyze proteins at Tyrosine, Tryptophan, and Phenylalanine residues with minimal Chymotrypsin autolysis byproducts or other protease contaminants. Immobilized Chymotrypsin resin is provided in a 50% slurry, and is supplied as 2 mL of actual resin (4 mL of slurry).
ProteoChem's Immobilized Chymotrypsin gel is manufactured from sequencing grade Chymotrypsin, which has been TLCK treated to remove tryptic activity. Immobilized Chymotrypsin is covalently bound to agarose, which allows for easy removal of the enzyme from digested sample. Recommended reaction conditions are pH 7.5 to 9.0 at 37ºC.

7. Immobilized Trypsin
ProteoChem’s highly active Immobilized Trypsin resin (Trypsin Agarose) is used to cleave proteins at the carboxyl side of Lys and Arg residues with minimal trypsin autolysis byproducts or other protease contaminants. Immobilized Trypsin gel is TPCK treated to remove chymotryptic activity. Recommended reaction conditions are pH 7.5 to 9.0 at 37ºC.
8. Immobilized Streptavidin
Immobilized Streptavidin is a chromatography resin with streptavidin covalently bound to 6% crosslinked beaded agarose. Immobilized Streptavidin resin is suitable for gravity flow columns, spin columns, and FPLC methods. ProteoChem's Immobilized Streptavidin resin is stable from pH 2-11 and is resistant to leaching of the streptavidin. The bead size is 45 to 165 um.
Immobilized Streptavidin can be used for affinity chromatography purifications by separating biotinylated proteins from non-biotinylated proteins and for affinity purification of antigens when used with biotinylated antibodies. Immobilized Streptavidin agarose resin is provided as a 50% slurry in PBS storage buffer containing 0.02% sodium azide; 5 ml settled resin in 10 ml total volume.

9. Desalting Resin
Desalting Resin is a crosslinked dextran gel (Sephadex G-25*) which is used to desalt crosslinked or biotinylated proteins, labeled antibody, or perform buffer exchanges. The bead size is 100-300 μm and 1 gram swells to 4-6 mL gel. The fractionation range for globular proteins is 1,000 - 5,000 MW and for dextrans is 100 - 5,000 MW.
